Additionally, Dyclonine hydrochloride effectively inhibits sodium fluorescein uptake in OATP1B3 and OATP1B1-transfected CHO cells, with inhibition rates of 83.62% and 170.92%, respectively, at 10 uM. In terms of antiviral activity, Dyclonine hydrochloride inhibits cell viability in Vero E6 cells infected with SARS-CoV-2, showing an inhibition index of 1.119 relative to the arbidol control. It also inhibits cytotoxicity in Caco-2 and Vero-6 cells exposed to SARS-CoV-2 at 10 µM with inhibition rates of -3.77% and -0.08%, respectively. However, it showed a negative inhibition percentage of -2.252% in the SARS-CoV-2 3CL-Pro protease inhibition assay at 20 µM concentration.
