研究表明生物体受刺激或扰动后(如低氧、营养物质、药物等)代谢产物会发生变化,基于肿瘤细胞内、外相关特征性小分子代谢标记物的异常变化来诊断恶性肿瘤、进行药物筛选、评价药物毒性等的方法也逐渐成为研究热点。癌细胞具有一种独特的、有别于正常细胞的代谢表型,监测癌细胞代谢过程中小分子代谢物的波动情况,将有利于预测肿瘤的进展、了解体内物质代谢途径、探索癌症发病机制及药物作用机制等。目前的研究主要集中于与癌细胞能量代谢相关的化合物研究,如核苷类、氨基酸类、脂类、糖类等。例如,已发现多种与肿瘤相关的脂类标记物, 胆碱、磷酸胆碱、磷酸卵磷脂、胆固醇等脂类变化谱是细胞膜破坏的标志;丙酮酸、乳酸、异丁酸等是与肿瘤细胞糖代谢相关的生物标记物, 探索通过靶向脂代谢、糖代谢途径治疗恶性肿瘤的策略正备受关注。
