体外活性 | 采用Ellman法评估Ethopropazine Hydrochloride对胆碱酯酶的抑制活性,以人重组乙酰胆碱酯酶(hAChE)和人血浆丁酰胆碱酯酶(hBChE)为靶酶,乙酰硫代胆碱(ATChCl)作为底物。在96孔板中,依次加入酶溶液、显色剂DTNB和不同浓度的Ethopropazine Hydrochloride(10mM-1nM),最后加入1mM ATChCl启动反应,反应在37℃下孵育5分钟后,于412nm波长下测定吸光度变化。结果表明,Ethopropazine Hydrochloride对hBChE表现出强效抑制(IC₅₀=1.6μM),而对hAChE的抑制作用较弱(IC₅₀=1020μM),是一种高效且高选择性的BChE抑制剂[1]。 |
