ASB 14780 is a 4-phenoxy derivative that exhibits highly selective inhibition of cytosolic phospholipase A2α (cPLA2α), with an IC₅₀ value as low as 20 nM. By inhibiting the production of pro-inflammatory mediators such as arachidonic acid, ASB 14780 effectively reduces TPA-induced ear edema and ovalbumin-induced asthma responses in vivo.

体内活性

ASB 14780（50 mg/kg；口服；单次给药）可抑制 TPA（tetradecanoyl phorbol acetate）在小鼠耳部引起耳壁增厚。[1]
ASB 14780（5 mg/kg；20 mg/kg；口服；每日一次）可减轻气道高反应性（AHR），并以剂量依赖性方式抑制即刻哮喘反应（IAR）和迟发哮喘反应（LAR）。[1]
根据口服给药（10 mg/kg；口服）与静脉注射给药（1 mg/kg；静脉注射）的 AUC 比值计算，ASB 14780 在小鼠体内表现出 89.6% 的良好口服生物利用度。[1]
