1. Tissue/cell: human rhinopolypus tissue; 4% Paraformaldehyde-fixed and paraffin-embedded;
Antigen retrieval: citrate buffer (0.01M, pH6.0), Boiling bathing for 15 min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30 min; Blocking buffer (normal goat serum) at 37°C for 20 min;
Incubation: Anti-IL-23 Polyclonal Antibody, Unconjugated (TMAB-00956) 1:300, overnight at 4°C, followed by conjugation to the secondary antibody and DAb staining.
2. Sample:
Lane 1: Mouse Ear tissue lysates
Lane 2: Human A431 cell lysates
Lane 3: Human Jurkat cell lysates
Primary: Anti-IL-23 (TMAB-00956) at 1/1000 dilution
Secondary: IRDye800CW Goat Anti-Rabbit IgG at 1/20000 dilution

研究背景IL-23 is a heterodimeric cytokine composed of two subunits: a unique p19 subunit, and a p40 subunit that is shared with IL-12. IL-23 is secreted by activated dendritic cells and macrophages. IL-23 binds to a heterodimeric receptor complex composed of IL12RB1 and IL23R, activates the Jak-Stat signaling cascade, stimulates memory rather than naive T cells and promotes production of proinflammatory cytokines, such as IFNγ and IL-17. IL-23 induces autoimmune inflammation and thus may be responsible for autoimmune inflammatory diseases and may be important for tumorigenesis. This recombinant human IL-23 is produced by human cells.(Tag free) Biological activity: The activity was measured by its ability to induce the IL-17 expression in murine splenocytes. Reconstitution: Briefly centrifuge the vial before opening. It is recommended to reconstitute the protein in sterile PBS containing 0.1% endotoxin-free recombinant human serum albumin.
