| 体外活性 | 通过荧光化合物N-acetyl-asp-glu-val-asp-7-amino-4-trifluoromethyl coumarin (Ac-DEVD-AFC)测量半胱天冬酶活性。研究表明,该底物快速进入细胞,在特定半胱天冬酶作用下有效地在天冬酰氨酸残基处被切割,产生荧光化合物7-amino-4-trifluoromethyl coumarin (AFC)。细胞破裂后,通过HPLC分离释放的AFC,并通过荧光检测。使用泛半胱天冬酶抑制剂benzyloxycarbonyl-val-ala-asp-fluoromethylketone可以阻断AFC在细胞中的出现,从而证实了细胞内半胱天冬酶负责该切割反应。在与多柔比星或放线霉素孵育约2小时后,首次观察到半胱天冬酶活性,之后AFC的产生随时间线性增长[1]。 |
