B cell maturation antigen (BCMA) is a member of the TNF receptor superfamily. It has been designated TNFRSF17. Mouse BCMA is a 185 amino acid (aa) protein consisting of a 49 aa extracellular domain, a 23 aa transmembrane domain, and a 113 aa intracellular domain. BCMA is a type III membrane protein containing one extracellular cysteine rich domain. Within the TNFRSF, it shares the highest homology with TACI. BCMA and TACI have both been shown to bind to APRIL and BAFF, members of the TNF ligand superfamily. BCMA expression has been found in immune organs and mature B cell lines. Although some expression has been observed at the cell surface, BCMA appears to be localized to the Golgi compartment. The binding of BCMA to APRIL or BAFF has been shown to stimulate IgM production in peripheral blood B cells and increase the survival of cultured B cells. This data suggests that BCMA may play an important role in B cell development,function and regulation.

常见问题解答

问如何稀释工作液？

答根据您的工作液浓度，计算母液所需要稀释的倍数和所需母液的体积。建议用水、生理盐水、PBS等溶剂进行稀释，如果是细胞实验，可以使用培养基。取适当的溶剂缓慢加入到母液中，直到达到您的工作液浓度，可以通过涡旋或者移液枪轻轻吹打帮助混匀。我们的化合物大部分是脂溶性的，所以在使用 PBS 或者培养基等稀释工作液的时候，会有沉淀析出，这些都是正常现象，大多时候可以通过超声完全溶解。工作液建议现配现用！

问是否能直接用缓冲液对抑制剂 DMSO 母液做梯度稀释？

答大部分情况下，都是可以溶解的。但有时，有机试剂直接加入水相介质时会析出。建议将抑制剂先以DMSO做梯度稀释，再将经过稀释的抑制剂加入缓冲液或细胞培养基。有些抑制剂甚至只有在其工作浓度下才能溶于水相。比如细胞实验中希望终浓度为 1 μM 的话，可以把 10 mM 的 DMSO 母液用 DMSO 稀释到 1 mM，再吸 2 μL 加入到 2 mL 的生理盐水/PBS/细胞培液，终浓度即为 1 μM。为避免药物析出，稀释前，可将母液和培养基 37℃ 预热，避免温度低造成严重析出。若稀释过程中出现化合物析出的情况，建议您采用超声加热的方法使其复溶。

问如果低于说明书给出的最高溶解度情况下，产品仍未溶解怎么办？

答建议您使用一些辅助溶解方法，如水浴加热至 45℃ 或用超声震荡，加速粉末溶解。

问Tween-20 可以用 Tween-80 替代吗？

答不建议使用 Tween-20。相较于 Tween-20，Tween-80 的耐受性更好，Tween-80已被用作评估实验药物和毒物的行为影响的工具，且没有明显的副作用。参考文献：Castro CA， Hogan JB， Benson KA， Shehata CW， Landauer MR. Behavioral effects of vehicles: DMSO， ethanol， Tween-20， Tween-80， and emulphor-620. Pharmacol Biochem Behav. 1995 Apr;50(4):521-6.

问PEG300 可以用 PEG400 替代吗？

答PEG300 具有适度的粘稠度，有利于药物的溶解和给药过程，同时也能够提供良好的耐受性，不会对动物造成不良影响。PEG400 可以替代 PEG300。 PEG600 不是很建议，因为熔点接近常温。

存储	Lyophilized powders can be stably stored for over 12 months, while liquid products can be stored for 6-12 months at -80°C. For reconstituted protein solutions, the solution can be stored at -20°C to -80°C for at least 3 months. Please avoid multiple freeze-thaw cycles and store products in aliquots.
