产品描述	Golimumab (CNTO-148) is a potent humanized anti-TNFα IgG1κ monoclonal antibody.Golimumab has anti-inflammatory and anticancer activity, inhibits the production of IL-6 and IL-1β, and induces apoptosis.Golimumab can be used to prevent inflammation and destruction of cartilage or bone, and can be used to study rheumatoid arthritis and cancer. 体外活性	Golimumab (CNTO-148) (0.1-10 μg/mL; 24-72小时; 跨膜TNFα-转染Jurkat细胞) 通过减少跨膜TNFα表达细胞的活性[1]。同时，在(10 μg/mL; 48小时; 跨膜TNFα-转染Jurkat细胞)的条件下，Golimumab (CNTO-148) 诱导凋亡，提高活性caspase-3水平，并促进凋亡性DNA碎片化[1]。
体内活性	Golimumab (CNTO-148) 在Tg197转基因小鼠模型中通过腹膜注射，每日剂量1-10 mg/kg，连续4周的治疗，能够缓解TNFα诱导的关节炎，模仿人类小鼠模型中的症状[3]。此外，通过静脉注射Golimumab (CNTO-148)，在健康的瑞士白色雄性小鼠上，每日剂量为24 mg/kg，持续7天的治疗，能够抑制氧化应激、阻止细胞凋亡和炎症反应，从而改善肾功能。Golimumab 减少了所有的肾损伤标志物并减轻了细胞死亡[3]。
