Tissue/cell: rat brain tissue; 4% Paraformaldehyde-fixed and paraffin-embedded;
Antigen retrieval: citrate buffer (0.01M, pH6.0), Boiling bathing for 15 min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30 min; Blocking buffer (normal goat serum) at 37°C for 20 min;
Incubation: Anti-VIP Polyclonal Antibody, Unconjugated (TMAB-01969) 1:200, overnight at 4°C, followed by conjugation to the secondary antibody and DAb staining.

研究背景VIP(Vasoactive Intestinal peptide) is a 28-amino acid peptide structurally related tosecretin. It was originally isolated from intestinal extracts and shown to be a potent vasodilator. Subsequent work demonstrated that VIP is very widely distributed in the peripheral and central nervous systems, and probably should not be considered a true GI hormone. A huge number of biological effects have been attributed to VIP. With respect to the digestive system, VIP seems to induce smooth muscle relaxation (lower esophageal sphincter, stomach, gallbladder), stimulate secretion of water into pancreatic juice and bile, and cause inhibition of gastric acid secretion and absorption from the intestinal lumen.
