1. Tissue/cell: rat lung tissue; 4% Paraformaldehyde-fixed and paraffin-embedded;
Antigen retrieval: citrate buffer (0.01 M, pH 6.0), Boiling bathing for 15 min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30 min; Blocking buffer (normal goat serum) at 37℃ for 20 min;
Incubation: Anti-RNASE4/Angiogenin Polyclonal Antibody, Unconjugated (TMAB-12274) 1: 200, overnight at 4°C, followed by conjugation to the secondary antibody and DAB staining
2. Sample:
Lane 1: Human HepG2 cell lysates
Lane 2: Human HUVEC cell lysates
Primary: Anti-RNASE4 (TMAB-12274) at 1/1000 dilution
Secondary: IRDye800CW Goat Anti-Rabbit IgG at 1/20000 dilution
Predicted band size: 14 kDa
Observed band size: 15 kDa

研究背景The protein encoded by this gene belongs to the pancreatic ribonuclease family. It plays an important role in mRNA cleavage and has marked specificity towards the 3' side of uridine nucleotides. Alternative splicing results in four transcript variants encoding the same protein. This gene and the gene that encodes angiogenin share promoters and 5' exons. Each gene splices to a unique downstream exon that contains its complete coding region. [provided by RefSeq, Aug 2013].
