1. Paraformaldehyde-fixed, paraffin embedded (Mouse liver); Antigen retrieval by microwave in sodium citrate buffer (pH6.0); Block endogenous peroxidase by 3% hydrogen peroxide for 30 minutes; Blocking buffer (3% BSA) at RT for 30 min; Antibody incubation with (HGF) Polyclonal Antibody, Unconjugated (TMAB-00852) at 1:400 overnight at 4°C, followed by conjugation to the secondary antibody (labeled with HRP) and DAB staining.

Lane 1: Rat Placenta tissue lysates
Lane 2: Rat Cerebellum tissue lysates
Primary: Anti-HGF (TMAB-00852) at 1/200 dilution
Secondary: IRDye800CW Goat Anti-Rabbit IgG at 1/20000 dilution

研究背景HGF is a pleiotropic protein in the plasminogen subfamily of S1 peptidases. HGF is a potent, mesenchymally-derived mitogen for mature parenchymal hepatocytes, and acts as a growth factor for a broad spectrum of tissues and cell types. HGF signals through a transmembrane tyrosine kinase receptor known as MET. Human HGF is secreted as an inactive 728 amino acid (aa) single chain propeptide. HGF regulates many physiological processes including proliferation, scattering, morphogenesis and survival. HGF is a crucial mitogen for liver regeneration processes, especially after partial hepatectomy and other liver injuries. This recombinant human HGF is produced by human cells. (Tag free) Biological activity: The activity was measured by its ability to stimulate the proliferation of the monkey epithelial cell line 4MBr-5. Reconstitution: Briefly centrifuge the vial before opening. It is recommended to reconstitute the protein in sterile PBS containing 0.1% endotoxin-free recombinant human serum albumin.
