首先,从ChEMBL数据库中获取了一组已知转录相关蛋白调节剂的参考集合(包含超过46,990个经过预筛选的化合物,涵盖逾120,000个活性数据条目)。接着,以Tanimoto指数≥0.8为条件(每个参考化合物最多限10个类似物),对内部数据库进行二维指纹相似性搜索,最终获得近23,700个转录相关分子(该扩展化合物集可根据需求提供)。随后对报道的活性值进行筛选(<10μM),并应用内部药物化学过滤器(排除PAINS结构、毒性及反应性化合物),最终得到包含超过9,200个类药筛选化合物的终版集合。
