化合物筛选
首先,从CHEMBL数据库获取了包含23,174个具有已知连接酶抑制活性的参考化合物集合。通过筛选保留仅对连接酶靶标具有中高活性的化合物,进一步过滤后得到包含10,000个独特连接酶调节剂的12,000条活性数据。
随后,以参考集合为基础对内部数据库进行二维指纹相似性搜索(塔尼莫托系数>0.85,每个参考化合物最多匹配50个类似物)。此外,最终筛选库中移除了含高反应性基团的分子,使其成为针对连接酶药物研发项目的优质新化学结构来源。
