重组蛋白产品该如何保存?
1) 存储条件:重组蛋白产品收到后,如不立即使用,应当保存在 -20℃ 至 -80℃ 的条件下。冻干粉可稳定保存 12 个月以上,液体类产品在 -80℃ 条件下可保存 6-12 个月。已复溶的重组蛋白样品需尽快用完,如果不能在一周内用完,需在 -20℃ 至 -80℃ 保存,可保存 3 个月以上\n2) 避免反复融冻:重组蛋白产品应注意避免反复冻融,建议将重组蛋白产品分装成多份。分装冻存时,建议选择含有一定浓度载体蛋白的溶液或培养基复溶产品。产品浓度不低于 100 ug/mL,且每管的体积不小于 20 uL。
重组蛋白产品是如何运输的?
大部分重组蛋白产品为冻⼲粉,对温度不敏感,在室温下稳定,一般室温或蓝冰运输。对于液体类蛋白,一般采用⼲冰运输。如果您收到产品时发现冰盒已经融化,无需担心,蛋白可以正常使用,短时间内不会影响产品质量。如果特定蛋白在室温下不稳定,我们会提供相应的低温运输方式,以确保产品质量不受影响。
蛋白能保证活性吗?
我们有现货的产品,有活性数据的话会展示在官网生物活性那栏,没有数据的也不代表就一定没有活性,是我们还没有建立活性检测方法,没有进行测试,但是我们的蛋白都是优先按照有活性的方法去表达纯化的。
为什么某些重组蛋白溶液中还需要添加载体蛋白?
载体蛋白通常被用于提高蛋白的稳定性,防止蛋白在冷冻或解冻过程中粘附在管壁上。塑料管对蛋白有一定的吸附能力,这可能导致蛋白和管壁之间难以分离,导致溶液中蛋白的实际浓度下降,从而影响其活性。为减少这种损失,在长期保存重组蛋白 产品之前,建议添加常用的载体蛋白溶液,例如0.1% BSA(牛血清白蛋白)、5% HSA(人血清白蛋白)、10% FBS(胎牛血清)或 5% 海藻糖等。如果重组蛋白的浓度较低,强烈建议添加适量的载体蛋白。
为什么重组蛋白的体积和质量与预期不符?
蛋白冻干粉由蛋白原液冻干而来,冻干粉质量不等同于蛋白质量。由于冻⼲产品的体积大⼩受多种因素影响,包括冻⼲前缓冲液的组分、盐离⼦浓度以及产品本身的浓度等。冻⼲粉可能看上去很多,也可能肉眼不易观察到,这些都是正常现象,请您放心使用。在收到产品后,照常复溶、使用或分装储存即可。

A DNA sequence encoding the human AA2AR (P29274) (Pro2-Leu208 & Glu219-Ala316) (TM domains 1-7) was expressed and linked by a Fusion Protein, with a Flag tag at the N-terminus and a polyhistidine tag at the C-terminus. Nanodisc is a versatile tool for studying membrane proteins. Using styrene-maleic acid (SMA) copolymer, membrane proteins can be extracted directly from prokaryotic and eukaryotic expression systems in the absence of detergents to preserve the protein structure and function better. Compared to membrane scaffold proteins (MSPs) nanodiscs, SMA nanodiscs also have the advantage of preserving proteins' nature by maintaining native lipids surrounded without introducing any heterologous proteins, which allows studies of protein structure and functions in a native-like environment.

研究背景
Adenosine Receptor Subtype A2a (A2aR) is a G-protein coupled receptor that mediates adenosine (a potent immunosuppressor) signaling in leukocytes. A2aR has been implicated in neuroinflammation and synaptic plasticity-related processes. Signaling through A2aRs inhibits local and systemic inflammation and A2aR expression showed protective effects against non-alcoholic steatohepatitis (NASH) in the liver. A2aR deficiency triggers spontaneous inflammation and NASH-Hepatocellular Carcinoma (HCC) in mice while low A2aR gene expression in human HCC is associated with hepatic inflammation and poor survival. Reducing adenosine signaling via A2aR antagonism provides an alternative targeted strategy to modulate the number and efficacy of infiltrating effector T cells within the immunosuppressive tumor microenvironment. Co-blockade of A2aR signaling and CD73 reduced tumor initiation, growth, and metastasis.
