1. Western blot analysis of RAD18 on different lysates using anti-RAD18 antibody at 1/1,000 dilution. Positive control: Lane 1: NIH/3T3, Lane 2: 293T, Lane 3: MCF-7.
2. Immunohistochemical analysis of paraffin-embedded rat testis tissue using anti-RAD18 antibody. Counter stained with hematoxylin.
3. ICC staining RAD18 in Hela cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.
4. ICC staining RAD18 in NIH/3T3 cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

研究背景The RING-type zinc finger protein Rad18 is essential in post-replication repair of damaged DNA and contributes to the maintenance of genomic stability. Rad18 maintains chromosomal DNA with the Rad54-dependent DNA repair pathway and recruits ubiquitin-conjugating enzymes in the Rad6 pathway. Rad18 functions in gap-filling of a daughter strand on replication of damaged DNA and is localized in the nucleus.
