| 验证活性 | 1. Immunofluorescence staining of Human Galectin3 in Hela cells. Cells were fixed with 4% PFA, permeabilzed with 1% Triton X-100 in PBS, blocked with 10% serum, and incubated with Rabbit anti-Human Galectin3 monoclonal antibody (1:60) at 37°C 1 hour. Then cells were stained with the Alexa Fluor® 488-conjugated Goat Anti-rabbit IgG secondary antibody (green). Positive staining was localized to nuclear and cytoplasm. 2. Flow cytometric analysis of Human Galectin3 expression on HeLa cells. The cells were treated according to manufacturer’s manual (BD Pharmingen™ Cat. No. 554714), stained with purified anti-Human Galectin3, then a FITC-conjugated second step antibody. The fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact cells. 3. Anti-Galectin3 rabbit monoclonal antibody at 1:500 dilution. -Lane A: HeLa Whole Cell Lysate. -Lane B: SW480 Whole Cell Lysate. -Lane C: A431 Whole Cell lysate. -Lysates/proteins at 30 μg per lane. -Secondary -Goat Anti-Rabbit IgG (H+L)/HRP at 1/10000 dilution. -Developed using the ECL technique. -Performed under reducing conditions. -Predicted band size:26 kDa. -Observed band size:30 kDa. 4. NKX3-1 was immunoprecipitated using: -Lane A:0.5 mg mouse pancreas Whole Cell Lysate. -4 µL anti-NKX3-1 rabbit polyclonal antibody and 60 μg of Immunomagnetic beads Protein A/G. -Primary antibody: -Anti-NKX3-1 rabbit polyclonal antibody, at 1:100 dilution. -Secondary antibody: -Goat Anti-Rabbit IgG (H+L)/HRP at 1/10000 dilution. -Developed using the ECL technique. -Performed under reducing conditions. -Predicted band size: 27 kDa. -Observed band size:30 kDa |

| 研究背景 | LGALS3 (Galectin 3) is a Protein Coding gene. This gene encodes a member of the galectin family of carbohydrate-binding proteins. The encoded protein is characterized by an N-terminal proline-rich tandem repeat domain and a single C-terminal carbohydrate recognition domain. LGALS3 is a beta-galactoside-binding lectin and plays a role in numerous cellular functions including apoptosis, innate immunity, cell adhesion, and T-cell regulation. LGALS3 has an important role in tumor progression through inhibition of apoptosis. LGALS3 expression is associated with neoplastic transformation and with differentiation of monocytes to macrophages. Elevated expression of LGALS3 has been demonstrated in the synovium of rheumatoid arthritis (RA). Diseases associated with LGALS3 include Follicular Adenoma and Papillary Carcinoma. |
