| 验证活性 | 1. Flow cytometric analysis of Human ENO3 expression on HeLa cells. The cells were treated according to manufacturer’s manual (BD Pharmingen™ Cat. No. 554714), stained with purified anti-Human ENO3, then a FITC-conjugated second step antibody. The fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact cells. 2. Anti-ENO3 mouse monoclonal antibody at 1:500 dilution. -Lane A: HepG2 Whole Cell Lysate. -Lane B: Hela Whole Cell Lysate. -Lane C: MOLT4 Whole Cell Lysate. -Lane D: Raji Whole Cell lysate. -Lysates/proteins at 30 μg per lane. -Secondary -Goat Anti-Mouse IgG H&L (Dylight800) at 1/15000 dilution. -Developed using the Odyssey technique. -Performed under reducing conditions. -Predicted band size:47 kDa. -Observed band size:50 kDa. 3. Immunofluorescence staining of ENO3 in Hela cells. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS,blocked with 10% serum, and incubated with mouse anti-Human ENO3 monoclonal antibody (dilution ratio 1:60) at 4°C overnight. Then cells were stained with the Alexa Fluor?594-conjugated Goat Anti-mouse IgG secondary antibody (red). Positive staining was localized to Cytoplasm. |

| 研究背景 | ENO3 is one of the three enolase isoenzymes found in mammals. As a homodimer, ENO3 is found in skeletal muscle cells in the adult. A switch from alpha enolase to beta enolase occurs in muscle tissue during development in rodents. Mutations in ENO3 gene can be associated with metabolic myopathies that may result from decreased stability of the enzyme. Two transcripts have been identified for ENO3 gene that differ only in their 5' UTR. ENO3 may play a role in muscle development and regeneration. It appears to have a function in striated muscle development and regeneration. |
