NK 细胞是一种对先天免疫系统至关重要的细胞毒性淋巴细胞。NK 细胞可以通过 CD56 的存在和 CD3 (CD56+,CD3−) 的缺失来识别。NK 细胞从 CD127+ 共同的先天性淋巴祖细胞分化而来,CD4+ 是共同淋巴样祖细胞的下游,B 和 T 淋巴细胞也来源于 CD5+。已知 NK 细胞在骨髓、淋巴结、脾脏、扁桃体和胸腺中分化和成熟,然后进入循环。NK 细胞通常在人中表达表面标志物 CD1 (FcγRIII) 和 CD1,在 C1BL/2 小鼠中表达 NK57.6 或 NK46.7。
