背景:在稳定的环境条件下细胞内蛋白质的生成和分解处于动态平衡,当在体外从细胞和组织中提取蛋白时,细胞内的蛋白酶和磷酸酶就会被释放出来或被激活,导致目的蛋白分解或去修饰。因此在提取物中添加适当的蛋白酶、磷酸酶等抑制剂是防止蛋白降解和去修饰的有效方法。

TargetMol 磷酸酶抑制剂Cocktail II 是近年来发现的一批天然产物,很有潜力的磷酸酶抑制,可以高效抑制常见磷酸酶对于蛋白的去磷酸化作用。其中(-)-p-Bromotetramisole Oxalate 是强效不可逆的碱性磷酸酶抑制剂;Cantharidin 是可逆的丝氨酸/苏氨酸磷酸酶抑制剂;Microcystin LR 是可逆的蛋白磷酸酶1和2A(PP1和PP2A)抑制剂。

产品应用

在室温下解冻(如果DMSO在室温下融化较慢,可考虑25℃水浴融化),开盖前请低速离心一下,以便将黏附于管壁的液体甩至管底。然后在实验前以1:100(v / v)稀释至溶液样品(如细胞裂解液或组织提取液),短暂涡旋震荡加速溶解。应用于:WB、IP、Co-IP、pull-down、IF、IHC、kinase assay等。
